Research Interests
Establishment of a vascular system is a key requirement for the development and survival of the mammalian embryo. In the adult, deregulation of vascular growth contributes to the pathology of devastating disorders including, but not restricted to cancer. We are interested in understanding the process by which blood vessels are formed and its regulation in vivo. Specifically, our research efforts focus on four major projects. a) Angiogenesis Inhibitors: Using a combination of transgenic mouse models, biochemistry and cell biology we investigate the biological function of thrombospondin-1, ADAMTS1 and ADAMTS8, all potential endogenous inhibitors of angiogenesis. Experiments are also designed to ascertain their potential threapeutic value in the suppression of tumor growth. b) Contribution of Notch1 to vascular morphogenesis: Recent linkage and knock-out studies have revealed that Notch signaling is required for morphogenesis and homeostasis of the vascular system. Using a Cre-lox strategy we are inactivating Notch1 from endothelial and smooth muscle cells. These experiments are complementary to expression arrays and several modalities of signaling studies in vitro. c) Regulation of vascular endothelial growth factor availability: We have recently found that specific proteases part take in the removal of VEGF from the matrix by cleaving the growth factor directly and releasing bioactive peptides. This release is dose-dependent and it is suppressed by specific inhibitors. Proteolytically processed VEGF is able to phosphorylate its receptors at the same molar ratio as the entire molecule. The biological significance of these findings is being studied with transgenic mice expressing different VEGF mutants. d) Role of progesterone receptor signaling in endothelial cells: Steroid hormones are essential for endometrial cycle, ovulation, implantation, and lactation. In addition they play significant roles in the cardiovascular system, yet relatively little is known about their effects on vascular cells. We are focusing on the role of progesterone receptor on endothelial cell biology using a combination of transgenic mouse models and signaling experiments in vitro.
Biography

Luisa Iruela-Arispe is a cell and developmental biologist and professor in the Department of Molecular, Cell and Developmental Biology. She earned her Ph.D. degree from the University of Sao Paulo in Brazil in 1989 and received post-doctoral training at the Universtity of Washington in Seattle. She was appointed Assistant Professor at the Department of Pathology at Harvard Medical School in 1994 where she stayed for four years. In 1998, Dr. Iruela-Arispe joined the faculty of the Department of Molecular, Cell and Developmental Biology at UCLA. Her research centers in understanding the molecular regulation of endothelial cell commitment and differentiation, patterning of the vasculature and homeostatic vascular function. This information has provided the basis to develop strategies to control vascular growth during disease, particularly in cancer. Dr. Iruela-Arispe is strongly committed to education. She is currently the Vice-Chair for teaching in her Department and provides instruction to both undergraduates and graduate students. She is actively involved in her field of research and has organized several meetings including two Gordon Conferences, one on Angiogenesis and a second on Vascular Biology. She is the founder and director of the Vascular Biology Training Program at UCLA.
